Federico amplified the upstream region of the gene called hydropsin5 in Matt Harms' undergraduate thesis.

We chose this gene because it has an element similar to a Pax-6 binding domain, which could be involved in directing opsin expression (a hypothesis that is not tested, just suggested based on sequence similarity).

In December 2011, THO tried to use Hydra gene gun technique at Irvine to test the plasmid. There was a promising result in 1 animal, with what appeared to be GFP expression.

However, the dsRed positive control never showed up, so this test was inconclusive and needs to be repeated. The technique can be repeated at UCSB using Joel Rothman's gene gun.
